重亚硫酸盐甲基化DNA分析处理试剂盒

甲基化特异性PCR(methylation-specific PCR, MSP)试剂盒
甲基化特异性的PCR是一种灵敏度高且操作相对简单的甲基化研究方法。重亚硫酸盐处理DNA后,基因组DNA发生的由甲基化状态决定的序列改变,随后进行引物特异性的PCR。该方法引物设计是关键。MSP中设计两对引物,即一对结合处理后的甲基化DNA链(引物对M),另一对结合处理后的非甲基化DNA链(引物对U)。检测MSP扩增产物,如果用引物M能扩增出片段,则说明检测位点存在甲基化;若用引物U扩增出片段,则说明被检测的位点不存在甲基化。Millipore公司的CpG WIZ ®甲基化特异性PCR试剂盒,提供了目的基因的甲基化和非甲基化引物以及被验证的PCR条件,同时提供对照DNA。

Millipore的EZ-TFA转录因子分析试剂盒,结合了EMSA和ELISA的原理,采用显色或化学发光检测,比同位素的方法更快,更容易,更灵敏,更安全,试剂盒检测原理:1. 生物素标记的具有目标转录因子结合保守序列的双链寡核苷酸与细胞/核提取物和TFA实验缓冲液混合孵育;2. 孵育期间,细胞/核提取物中有活性的目标转录因子将结合寡核苷酸保守序列;3. 混合物转移到亲和素包被的96孔上有活性的目标转绿因子通过生物素标记的寡核苷酸被固定在板子上;4. 无活性的未结合的转录因子被洗去;5. 目标转录因子的活性通过抗体、HRP标记二抗和底物反应经化学发光或显色反应被检测。
